1110 Bioassay-guided fractions of garcinia kola nuts inhibit cell proliferation via modulation of PI3K/Akt/mTOR pathway in melanoma and non-melanoma skin cancers

Melanoma (MSC) and non-melanoma skin cancer (NMSC) are the most common cancers in United States. Although NMSC accounts for greater proportion of diagnosed cases, MSC is more aggressive responsible over 80% cancer-related deaths. Adverse effects and/or resistance associated with existing treatment regimens have prompted new research efforts directed toward finding safer efficacious chemopreventive- and- therapeutic strategies to curb incidence mortality. Natural products reliable sources potentially safe effective agents. Garcinia kola a phenolic-rich herb grown west Africa. The nut, commonly called Bitter Kola, eaten recreationally used as traditional medicine several diseases, including hepatoprotective, bronchodilating, antidiabetic, antioxidant activities. In this study, three melanoma (A375, SK-Mel-28, B16F10), (A431, SCC12-12, UW-BCC1), normal (NTERT Keratinocytes) cell lines were treated 100-400ug methanolic extract (MeOH), N-hexane (Hex) faction Hexane 75%-Ethyl acetate 25% subfraction (F3) evaluate their on proliferation, apoptosis, PI3K/Akt/mTOR pathway signaling. MeOH Hex treatments resulted (i) significant dose-dependent decrease growth viability (ii) increased expression apoptotic markers (iii) downregulation pathway.
